Abstract Corynebacterium spp. and Erysipelothrix rhusiopathiae susceptibility test is currently performed using 2.5-5.0%v/v lysed horse blood at 35°C as an adjuvant to Mueller-Hinton broth, which is recommended by the Clinical and Laboratory Standards Institute(CLSI). We evaluated the usefulness of Strepto-Haemo Supplement "Eiken®" (SHS) contains 1.5 ml of lysed horse blood, 1.07 mg of beta-NAD, and 0.36 g of yeast extract, compared with the current standard lysed horse blood, as an adjuvant to Mueller-Hinton broth for the Corynebacterium spp. and E. rhusiopathiae susceptibility test. For all 58 clinical isolates of Corynebacterium spp. and E. rhusiopathiae tested, the SHS method and the standard CLSI method showed almost identical MIC results within one dilution difference. We believe that the SHS can be an alternative to the currently recommended lysed horse blood to test Corynebacterium spp. and E. rhusiopathiae susceptibility.
